Former FBI head James the former director has pleaded not guilty to giving untrue testimony to lawmakers and hindering a lawmaking procedure.
Mr Comey's lawyer submitted the response acting as his counsel in a national courtroom in the Virginia city of Alexandria on Wednesday.
Patrick Fitzgerald announced he would attempt to have the legal matter terminated for multiple factors including that his the accused, a opponent of President Donald Trump, was being targeted.
The former director was formally accused soon after Donald Trump pressured his chief prosecutor to proceed against him.
In court on mid-week, the defense counsel Mr Fitzgerald told the judge they aimed to file multiple motions to dismiss the legal matter before a court case, arguing the prosecution was retaliatory and that a federal prosecutor was wrongfully assigned to take over the proceedings.
James Comey's legal matter was initially handled by the original attorney, who left his position under scrutiny from the former president after his examination into another political adversary - New York Attorney General Letitia James - was unable to file indictments.
Mr Comey came across in optimistic frame of mind as he entered the courtroom on Wednesday, speaking with his attorneys and making jokes. He was supported by his partner, Mrs Failor and daughter his daughter, a government attorney who the previous administration terminated.
Following the reading to the magistrate recite his constitutional guarantees and the dual charges against him in the courtroom on mid-week, James Comey was inquired if he understood the accusations.
"Yes your honour. Appreciate it," he informed the proceedings.
James Comey was the head of the FBI from 2013-2017 and was dismissed about several months into Donald Trump's first term as national leader. At the time, he was overseeing an probe into Russian election interference and whether there were any relationships between Moscow and Trump's campaign.
While in office, Mr Comey sparked a counter-response from the Democratic party when he stated just days before the national vote that he was probing presidential nominee Clinton's use of a non-governmental email account. Indictments against the candidate were never brought, resulting in criticism from Republicans as well.
Prosecutors allege the former director provided false information to Congress during a Senate Judiciary Committee hearing in 2020 when he was being interrogated about both the email probe as well as the election interference investigation. They allege he provided false information to the upper house by stating he had not approved an individual at the agency to release to journalistic entities details about the government examinations.
Prosecutors also accuse James Comey of "improperly attempting to influence, obstruct and delay" the panel by providing untrue information to it.
Via social media Mr Comey published to his Instagram after he was charged, he said he was wrongly accused and alleged Trump of acting like a "tyrant".
"My family and I have been aware for years that there are consequences to challenging Mr Trump," he commented. "We refuse to live on our kneeling."
"I'm innocent," he stated further. "Thus initiate a legal proceeding."
The accusations against James Comey emerged after the former president posted on online platforms calling for his chief prosecutor, the attorney general, charge the former director and others.
